Maddam Sir revolves around a group of five police officers stationed in Lucknow, consisting of four women and one man. Together, they collaborate to solve various cases, and an unexpected turn of events occurs when one of the prisoners becomes a secret informant for them.
Among the female officers, Karishma and Haseena possess distinct personalities, leading to occasional conflicts between them. Nevertheless, they remain supportive of each other. A new officer named Anubhav joins their team and aids in resolving their differences.
Haseena develops romantic feelings for Anubhav, but later discovers that his affection was only a pretense for a mission. Despite this revelation, Anubhav's love for Haseena continues to grow, ultimately drawing them closer.
Anubhav and Haseena plan their engagement, but on the day of the ceremony, Anubhav receives an order to cancel the marriage and depart the city for a life-threatening national security mission.
