Phoenix Wright: Ace Attorney is a game where players become Phoenix Wright, a new lawyer. They defend their clients in five cases, which they can play in any order after finishing them once. Each case starts with a cinematic showing a murder, then Phoenix defends the suspect in court.

There are two parts to the game: investigations and courtroom trials. During investigations, players talk to characters to gather information and evidence. They can examine things in the environment and move to different places. Players use evidence to get new information from witnesses.

In the fifth case, players can examine evidence closely, rotating and zooming in on it using touchscreen controls. They can also use forensic tests to find clues at crime scenes. Players spray luminol to reveal hidden blood stains and apply aluminum powder to search for fingerprints. Then, they blow into the microphone to reveal the prints.

During courtroom trials, players try to prove their clients innocent by cross-examining witnesses. They look for lies and inconsistencies in the witnesses' testimonies and present evidence that contradicts them. Players can choose to press the witness for more details or present evidence by using vocal commands or swinging the Wii Remote. They also have to answer questions from the judge, witnesses, or prosecutor. If they make too many mistakes, their client is found guilty, and they have to start over. Winning a case unlocks a new one to play.

Phoenix Wright, a new defense attorney, helps his friend Larry Butz prove his innocence in the murder of Cindy Stone. They reveal the true culprit, Frank Sahwit.

"Turnabout Sisters": Mia Fey, Phoenix's mentor, is killed, and her sister Maya is arrested. Phoenix defends Maya against accusations and exposes Redd White as the true killer. Maya becomes Phoenix's assistant.
