In September 2018, during the promotion of Marvel's Spider-Man (2018), the game's creative director, Bryan Intihar, discussed the absence of Spider-Man's symbiote suit from the comics. He expressed that simply making it an unlockable suit wouldn't do justice to its significance. Intihar believed that the symbiote story is one of the best for Spider-Man and requires a proper telling in Insomniac's style.
Both Marvel's Spider-Man and its spin-off Marvel's Spider-Man: Miles Morales (2020) hinted at future developments in their post-credits scenes. These teases included the introduction of Venom, as well as featuring Norman Osborn and his son Harry in more prominent roles.
In June 2020, Insomniac announced Marvel's Spider-Man: Miles Morales as a standalone title but assured fans that they still had more of Peter's story to tell in future games.
Marvel's Spider-Man 2 and its companion game Marvel's Wolverine were jointly announced by Insomniac Games in September 2021. Bryan Intihar and Ryan Smith returned as creative and game directors, respectively. Yuri Lowenthal, Nadji Jeter, and Tony Todd were confirmed to star as Peter Parker, Miles Morales, and Venom.
According to Marvel Games vice president Bill Rosemann, the narrative of Spider-Man 2 would be darker and considered the next big chapter in the story. He compared the tone to The Empire Strikes Back, contrasting it with the first game's tonal similarity to the original Star Wars.
In January 2022, Brittney Morris, known for her work on the Miles Morales tie-in novel Wings of Fury, joined the writing team. Insomniac also brought on Davison Carvalho, a former concept artist for Marvel Cinematic Universe films, as the game's art director.
Actor Scott Porter, who previously voiced Harry Osborn in the first game's Research Station missions, revealed in November 2022 that he had been recast for Marvel's Spider-Man 2 to accommodate the character's expanded role. The decision was made to achieve a photorealistic portrayal and address any age gap concerns.
